Creative Biolabs provides the first rank affimer phage display library construction service for our clients all over the world. The affimer library is generated through our advanced Hi-AffiTM phage display platform, which can obtain higher affinity and diversity than that of other technology.

Affimer, an evolution of peptide aptamers, is a highly stable recombinant protein which can bind to specifically target protein and potentially interfere with its function under intracellular conditions. It is derived from the cysteine protease inhibitor family of cystatins and shares the common tertiary structure of an α-helix lying on top of an anti-parallel β-sheet. The protein is then engineered to display two peptide loops and an N-terminal sequence which can be randomized to bind different target proteins with high affinity and specificity similar to antibody. In addition, the engineered protein is stable, non-toxic, biological neutral and contain no post-translational modifications or disulphide bridges.

The Key Properties of Affimer:


Compared with common antibody (150-160 kDa), affimer (12-14 kDa) is ten-time smaller protein which can address similar function with antibody and even some applications where antibodies cannot be used. Affimer represents useful molecules for manipulating protein function in vivo, mostly as inhibitors of individual signaling components that prevent the interaction of the target protein with functionally important cellular partners. It also can be used as research tools across a number of platforms, such as ELISA, western blot, surface plasmon resonance (SPR), affinity purification, immunocytochemistry and flow cytometry. In addition, affimer libraries can be generated for screening in yeast two hybrid and phage display formats, extending their versatility. Consequently, these facts make affimer a valuable candidate for a wide range of applications in diagnostics, drug or biomarker discovery, life science research and therapeutics.

Based on conventional exogenous gene expression method through fusing the target genes to bacteriophage coat proteins then displaying on the phage surfaces to select specific binders, our Hi-AffiTM phage display platform has combined trimer codon technology and NNK method to introduce more diversity in library construction and further develop the affinity and diversity of generated libraries. This platform is more suitable for sorting and isolating the high affinity protein or peptide targets and ensure our affimer phage display library to achieve 100% precise mutant and over 1010 diversity.
