Relying on a simple process, electroporation emerges as a powerful tool for the genetic modification of diverse cell types based on the transient disruption of cell membrane via exposure to an electric field, which allows charged molecules to enter the cell. It can be used with most cell types, especially T cells, yielding a high frequency of both stable transformation and transient gene expression. Electroporation is easier to operate compared with the methods of virus-based transfection and transposons transfection. The main advantage of electric transfection is its applicability for transient and stable transfection of T cells. Moreover, electric transfection is easy and rapid to transfect a large number of T cells in a short time.
On the other hand, the main potential disadvantage of electroporation is the excessive cell death and the low transfection efficiency, which is varied with cell types and electroporation conditions (voltage, wave profile, time of the pulse, and buffer composition). Therefore, Creative Biolabs has optimized the process of electric transfection by distributing the electrical pulse equally among the cells and maintaining a stable pH and modifying the pulse and field strength parameters to reduce the loss as described above.
