T Cell Persistence in TCR-T Therapy

In the context of TCR-T therapy, T cell persistence refers to the ability of TCR-T cells to persist in the body after infusion. The persistence of TCR-T cells is important for their efficacy, as they need to remain in the body in sufficient numbers to continue targeting and killing cancer cells, leading to long-term antitumor activity. Good persistence of TCR-T cells can improve the effective antitumor response. Several factors can influence TCR-T cell persistence, including TCR-T cell designs and manufacturing processes, the type of TCR-T cell, the type and stage of the cancer being treated, and the patient's immune system.

  • Flow cytometry

Flow cytometry enables the identification of surface epitopes on live TCR T-cells. This method involves staining the TCR-T cells with fluorescent antibodies specific to the TCR molecule and other cell surface markers. The stained cells are then analyzed using flow cytometry to quantify the number of TCR-T cells present in a sample.

  • qPCR

qPCR is the gold standard for the detection and quantification of the integrated TCR transgene sequence in TCR-T cells. Monitoring TCR-T expansion/persistence through our qPCR assay gives you real-time data on the TCR-T therapy.

  • ddPCR

ddPCR is a powerful tool that can be used to determine the absolute number of TCR-T cells present in a blood sample. It enables the monitoring of TCR T-cell kinetics with high precision and sensitivity.
