Membrane proteins are proteins that are attached to or associated with the biological membrane. They act as transporters, channels, and receptors to enable information transmission and materials exchange across the plasma membrane. As such, they manifest as primary targets for cellular functioning and therapeutic agents. Cell-free Expression System

Due to the low abundance of membrane proteins in their natural hosts, it is necessary to overexpress these proteins in heterologous systems. Conventional cell-based systems can be problematic in insufficient membrane insertion, precipitation of de novo synthesized proteins, or even cytotoxicity due to the extensive alterations in the host cell's metabolism. Cell-free system, which is based on the modified lysates of various cellular components, provides powerful machinery for in vivo transcription/translation of target proteins independent of a living cell's integrity.
