The PD-1+TIGIT Combination Bioassay Kit is a bioluminescent cell-based assay that can be used to measure the potency and stability of antibodies and other biologics targeting PD-1 and TIGIT.

Molecular Mechanism of ActionThe Bioassay Kit consists of two genetically engineered cell lines, PD-1+TIGIT Effector Cells and PD-L1+CD155 aAPC/CHO-K1 Cells. When co-cultured, PD-1 inhibits TCR pathway-activated luminescence, and TIGIT inhibits CD226 pathway-activated luminescence. The addition of anti-PD-1 Ab blocks PD-1 binding to PD-L1, resulting in full TCR pathway activation. The addition of anti-TIGIT Ab blocks the TIGIT/CD155 interaction, thereby re-establishing CD226 pathway-activated luminescence. Blocking of PD-1/PD-L1 and TIGIT/CD155 can be detected in a dose-dependent manner by addition of Bio-Glo Reagent and quantitation with a luminometer.

Target/DescriptionPD-1 and TIGIT are both immune inhibitory receptors that are expressed on T cells. Engagement of PD-1 by PD-L1 on an adjacent cell inhibits TCR signaling and TCR-mediated proliferation, transcriptional activation and cytokine production. The ligand for TIGIT is CD155. TIGIT negatively regulates NK cell killing and T cell activation by competing with CD226 for binding to CD155 and by directly interfering with CD226 homodimerization.

CompatibilityEach kit contains sufficient reagents for 600 assays using the inner 60 wells of ten 96-well plates which can do 20 antibody analysis (10 concentration dose*3 repeats).
Format96- and 384-well plate formats
StorageUpon arrival, immediately transfer the cell vials to below -140°C (freezer or liquid nitrogen vapor phase) for long-term storage. Do not store cell vials submerged in liquid nitrogen. Do not store cell vials at -80°C as this will negatively affect cell viability and performance.
Store Bio-Glo Luciferase Assay Buffer, Bio-Glo Luciferase Assay Substrate and Fetal Bovine Serum at -20°C. Avoid multiple freeze-thaw cycles of the serum.
We recommend preparing Bio-Glo Reagent immediately before use. Do not store and use the reconstituted reagent.
Store RPMI 1640 Medium at 4°C protected from fluorescent light.
