In the antibody research, chicken is considered an ideal host for generating antibodies against mammalian or human protein antigens. Since chickens are at least 100 million years removed from mammals, they tend to recognize any mammalian gene product as foreign, and mount vigorous immune responses. In addition, a faster antibody response can occur in chicken hosts compared to in mammalian species. Moreover, the process of producing chicken antibodies is also much easier compared to other host species, as the specific antibodies are generally isolated from the egg yolk.

The distinct advantages for using chickens produce antibodies are including:

Higher titre.
More stable than the other mammalian antibodies. The chicken antibody has shelf-lives of 5 years or more at 4˚C.
Much easier to perform double immunostaining. Secondary antibodies against chicken IgY’s don’t cross react with mammalian IgG’s.
A larger glycosylation index, allowing more labeling with HRP and other antibody tags. This produces higher signals.
No mammalian “Fc domain” within the chicken IgY. Thus, it does not bind to mammalian Fc receptors, avoiding non-specific binding to cells expressing such receptors (e.g., macrophages and dendritic cells).

This platform is based on antigen-specific B lymphocytes flow cytometry technology, which can isolate antigen-specific B cells from bone marrow cells, lymph node cells or peripheral blood cells of immunized chicken by FACS.
