Unlike antibodies in humans or rodents, there are four types of immunoglobulin (Ig) found in rabbits, which are IgA, IgE, IgG, and IgM. There is only one isotype of IgG in rabbits, which is the primary type of antibodies used for extensive research and applications. Rabbit polyclonal antibodies have been applied in the diagnosis and treatment of a series of diseases. The application of rabbit monoclonal antibodies (mAbs) has also attracted increasing interests due to the capability of the unlimited supply of rabbit mAbs. A lot of effort has been put, in the past decades, in developing and making rabbit mAbs. This is particularly important for the antigens without immunogenicity in rodent animals. The rabbit antibody repertoire is an exceptional source for monoclonal antibodies (mAbs) that combine high specificity with high avidity and affinity. Thus, rabbit mAbs have higher dilution ratios and better labeling efficiency, in certain cases, without the requirement for antigen retrieval.

As rabbit antibodies often carry multiple glycans on the heavy chain Fc region (which cannot be removed by PNgase F), the intact mass determination is a general problem for rabbit antibodies (with or without BSA). However, our sequences are of such high quality that validation by intact mass is typically not required. Please also keep in mind that rabbit antibodies have an additional SH-bridge from the VL variable region to the VC region. This SH-bridge does not exist in mouse, hamster, rat or human antibodies, which add difficulties for the sequencing. Our DASS technology based sequencing service can solve this problem efficiently. In addition, customers have our coverage guarantee: each amino acid position will be covered by at least 20-70 different peptides.
