AOC for Electrochemical Proximity Assay (ECPA)

AOC Used for Electrochemical Proximity Assay (ECPA)

ECPA is a simple electrochemical form, which is based on the combination of the concept of proximity assay and electrochemical detection. ECPA produces a direct readout, which is suitable for high sensitivity (low femtomolar (FM) range) and selective quantification of various protein targets. ECPA consists of binding molecules (initially aptamers) and oligonucleotide (ON)-tags. Upon the formation of a cooperative complex with a protein target causing a redox-active moiety such as methylene blue closer to a gold electrode, it can produce a redox current. The current can be quantified with high precision and directly depends on the concentration of the target.

AOC used in the ECPA mainly utilizes the proximity effect of two AOC probes bound to a protein target to move the electrochemical active label-methylene blue (MB) closer to the gold electrode with preassembled thiolated DNA/competitor monolayer. In the presence of a protein target, the redox current in ECPA is quantitatively analyzed by square wave voltammetry (SWV). It is found that the redox current is directly dependent on the concentration of the target. ECPA can be used for any protein with available antibody pairs.
