C-Kit

KIT proto-oncogene receptor tyrosine kinase (C-Kit) is a mast cell growth factor (MGF) type 3 transmembrane receptor also known as KIT, PBT, SCFR, or CD117. It is a cell membrane Tyr protein kinase that belongs to the CSF-1/PDGF receptor subfamily of the protein kinase superfamily. C-KIT contains five Ig-like C2-type domains in the extracellular portion and one protein kinase domain in the cytosolic portion. Upon ligand (cytokine KITLG/SCF) binding, C-KIT is involved in many cellular pathways (e.g. AKT1 signaling pathway, GRB2 pathway via the activation of RAS, RAF1 and the MAP kinases MAPK1/ERK2 and/or MAPK3/ERK1, promoting STAT1, STAT3, STAT5A and STAT5B activation…) and it dictates the fate of cells by regulating cell survival and proliferation. It is also involved in regulating hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and melanogenesis.

C-KIT is associated with gastrointestinal stromal tumor, testicular germ cell tumor, leukemia, and acute myelogenous. The C-KIT signaling is mediated by protein phosphorylation and the rapid internalization and degradation of the receptors, making it an ideal target for ADC-based immunotherapeutics development.
