CD248

CD248, also named as tumor endothelial marker 1 or endosialin, is a single-pass transmembrane receptor. It belongs to the "Group XIV" family, which is a novel family of C-type lectin transmembrane receptors which just served a role in cell-cell adhesion processes but also in host defense. The ligands of CD248 are known to be extracellular matrix molecules (fibronectin and type I/IV collagen). CD248 is extensively expressed on mesenchymal cells in the developing embryo and also plays an essential role in proliferation and migration of pericytes and fibroblasts. Besides, expression of CD248 is notably decreased in adults, however, it enables to be up-regulated during inflammation, fibrosis, and malignancy.
Importantly, CD248 is essential for the progression of certain tumors. It has reported that abdominal tumors implanted into CD248-knockout (CD248−/−) mice have revealed a decrease in growth, invasiveness, and metastasis. Moreover, in human kidney disease, the level of CD248 expression is predictive of fibrosis and disease outcome. Additionally, it is highly expressed in the rheumatoid synovium, and it has been revealed to exacerbate inflammatory arthritis in mice, although the molecular basis of this phenotype has not been fully elucidated.
