CSF1R

Colony-stimulating factor 1 receptor (CSF1R) encoded by the c-fms proto-oncogene, is one of the most primary regulators in the immune system, which will further influence the development of the tumor. CSF-1 binds to CSF-1R and comes into being phosphorylation and kinase domain activation. Meanwhile, CSF-1R forms into homodimer. Tyrosine residues of the effect protein will be phosphorylated, which leads to macrophages' activation. The activated mononuclear scavenger system could promote the development of tumor through many pathways, such as promoting tumor growth, angiogenesis, extracellular matrix breakdown, invasion, and metastasis. Besides, studies have shown that CSF-1/ CSF-1R also affects tumor cells directly. Various malignant tumors have been confirmed CSF-1R overexpression.
The expression of CSF-1R in radiotherapy-resisted patients is 4.1 times higher than radiation sensitive cases, suggesting that CSF is an important factor to regulate and control tumor progression. The maturation and differentiation of tissue macrophages depend upon the activation of tightly regulated pathways. A number of studies have revealed that CSF1 binds to the high-affinity receptor tyrosine kinase, CSF1R. and is a key cytokine involved in the recruitment and activation of tissue macrophages. Aberrant expression of CSF1R mRNA has been described in cHL-derived cell lines. Since the CSF1/CSF1R pathway could be inhibited by small molecules. and some in vitro studies also suggest that CSF1R could be an interesting therapeutic target for cHL.
