TNF superfamily member 11(TNFSF11) is a member of the tumor necrosis factor cytokine family. It is a ligand for osteoprotegerin and acts as a key factor for osteoclast differentiation and activation. TNFSF11 is reported to be the dendritic cell survival factor and is associated with the regulation of T cell-dependent immune response. A number of studies have been revealed that T cell activation induces expression of TNFSF11 and leads to the increasing of osteoclastogenesis which is a rare disease characterized by abnormally dense bone. TNFSF11 induces osteoclastogenesis by activating multiple signaling pathways in osteoclast precursor cells. TNFSF11 activates anti-apoptotic kinase AKT/PKB through a signaling complex involving SRC kinase, a kinase that includes nine members. It also activates the tumor necrosis factor receptor-associated factor 6 (TRAF 6), indicating that TNFSF11 plays a role in the apoptotic regulation.
TNFSF11 is related to several diseases and disorders. Disruption of TNFSF11 in mice leads to severe osteopetrosis and a lack of osteoclasts. The deficient mice exhibited defects in early differentiation of T and B lymphocytes and cannot form lobulo-alveolar mammary structures during the pregnancy.
