Avimer, short for avidity multimer, is a kind of artificial multi-domain protein which is able to specifically bind certain antigens via multiple binding sites. This protein is also known as maxibody or low-density lipoprotein receptor (LDLR) domain A. It consists of two or more peptide sequences, which are based on A domains. The A domains are 30 to 35 amino acids scaffolds (~4 kDa) derived from extracellular cysteine-rich cell surface receptor proteins, and stabilized by disulfide bond formation and complexation of calcium ions. Each A domain can bind to a certain epitope of one target protein. When the combination of A domains binding to different epitopes of the same protein, the avimer will obtain increased affinity and avidity of this protein. Alternatively, if those A domains against epitopes on different target proteins, the avimer will be able to recognize multiple proteins, which is similar with bispecific antibodies. The construction of avimers with novel binding activities was achieved via phage display, yielding a set of different artificial domains and subsequent combination in one fusion polypeptide. Compared with common antibodies, avimers have improved heat stability but limited plasma half-life because of their smaller size. Since they are not structurally related to antibodies, avimers are classified as a type of antibody mimetic.
