Rosa26 Safe Harbor Locus
Inserting transgenes into a pre-selected locus with a single copy can overcome the defects of random integration. The safe harbor luci are well-known sites for safe gene integration without negative effects on the host cell. Rosa26 is a commonly used safe harbor locus in the mammalian genome. It was originally characterized in mice, as well as in humans, pigs, rats, and rabbits. Remarkably, ROSA26-inserted transgene is expressed in all tissues.
