Single-chain triplebody (Sctb) can be regarded as tandem scFv in terms of the structure. It has three tandem scFv units, bispecific or trispecific, 2+1 or 1+1+1 antigen-binding valency. In sctbs, there are two binding sites for antigens on the disease, such as tumor, cell surface and one for trigger molecule on the effector cell, which allow the recruitment of immune cells to eliminate disease cells, such as tumor cells. This type of BsAbs have been studied in tumor immunotherapy. The data from some studied showed that sctbs had increased redirected lysis function over the corresponding bsscFvs.
