Background

Designated as a homodimer primarily identified in monocytes, macrophages, and neutrophils, CLEC12A is considered a marker of primitive cells in acute myeloid leukemia. Its expression is subject to negative regulation under inflammatory stimulation and it counteracts positive regulatory signals by recruiting the tyrosine phosphatase SHP-1.

CLEC12A is capable of detecting monosodium urate crystals generated during the process of cell death. By binding with these crystals, CLEC12A serves as a preventative measure against harmful inflammatory reactions. Furthermore, CLEC12A restricts the recruitment of neutrophils in tissues following cell damage by inhibiting the production of CXCL1, CXCL10, and IL8. In essence, CLEC12A finely tunes immune responses, providing a feedback mechanism that is defensive to guard tissues from excessive inflammation-induced damage. Additionally, it is suggested that CLEC12A, in concert with the IFN response, has a participatory role in the immune processes that pressure the host's resistance to viral infection.
