Background

The FAS gene encodes the TNFSF6 receptor, also known as CD95, belonging to the TNF-receptor superfamily. Positioned on the cell surface, FAS is an integral component of the death-inducing signal complex (DISC). Within the DISC, the FAS-associated death domain protein recruits caspase-8 by binding to FAS, initiating downstream signaling that mediates apoptosis. This apoptotic process contributes to the antigen-stimulated suicide of mature T-cells and the induction of peripheral tolerance. Recent research highlights FAS as a tumor suppressor, revealing that the FasL ligand induces apoptosis in various types of cancer cells through the FAS receptor in cultured cells.
