β-Klotho (KLB) is a transmembrane protein with a molecular weight of 120 kDa, which is highly expressed in adipose tissue, liver, pancreas, and brain. KLB acts as a high-affinity receptor for FGF19 and FGF21 by binding to two highly conserved peptide regions in the C-terminal tail of them, thereby participating in a variety of metabolic processes in FGF21 and FGF19, such as glucose and lipid metabolism and bile acid synthesis. It has been reported that KLB may be involved in processing receptor complexes or proteasomal degradation in addition to its conventional role as a co-receptor. Additionally, KLB has been validated as a promising drug target for the treatment of various metabolic disorders, including nonalcoholic steatohepatitis (NASH), type 2 diabetes, and primary biliary cholangitis.
