Nkp30, an activating receptor of the natural cytotoxicity receptor (NCR) family, is a pivotal regulator of natural killer (NK) cell-mediated antitumor immunity. Structurally characterized by a single extracellular V-type immunoglobulin domain, Nkp30 associates with CD3ζ or FcεRIγ chains to transduce activation signals. It engages tumor ligands, including B7-H6 and stress-induced BAT3, triggering NK cytotoxicity against malignancies. Soluble β-glucan enhances Nkp30 clustering, rebalancing inhibitory signals from galectin-3 to promote tumor lysis. Beyond oncology, Nkp30's β-glucan recognition confers antifungal defense, while its expression in cytotoxic T-cell subsets expands immunotherapeutic potential. Emerging biotherapeutics, such as Nkp30-targeting multispecific antibodies, exploit these mechanisms for next-generation cancer therapies.
