Background

The complex glycosylated molecules, Thyroid Peroxidase (TPO) and Thyroglobulin (TG) are predominantly expressed in the thyroid. TG is the most voluminous and prevalent self-antigen of the thyroid. The iodized TG serves as the primary precursor for T4 or T3 hormone activity. Meanwhile, TPO is the central enzyme contributing to the production of thyroid hormones.

Beyond their role in thyroid physiology, TPO and TG are recognized as being associated with autoimmunity and lymphocytic infiltration of the thyroid, thus playing a critical role in the process of hypothyroidism. The susceptibility and therapeutic potential for autoimmune thyroid diseases, particularly Graves' disease, Hashimoto's Thyroiditis, and Hashimoto's encephalopathy, have been demonstrated by the presence of anti-TPO and/or TG antibodies.
