TIGIT (T cell immunoglobulin and ITIM domain) is an emerging immune checkpoint receptor co-expressed on NK cells and T cells, functioning as a critical regulator of antitumor immunity. Structurally characterized by an immunoglobulin variable domain and an immunoreceptor tyrosine-based inhibitory motif (ITIM), TIGIT binds with high affinity to poliovirus receptor (PVR) ligands overexpressed on tumor cells, transmitting inhibitory signals that suppress cytotoxic lymphocyte activity through competitive disruption of DNAM-1-mediated activating signals. This mechanism parallels PD-1/PD-L1 pathway modulation but operates through distinct molecular interactions within the tumor microenvironment.
