Characteristics of B cells

B cells or B lymphocytes arise from hematopoietic stem cells and serve as an important lymphocyte cell type in the mammalian adaptive immune system producing humoral immunity responses. B lymphocytes perform multiple functions, such as producing antibodies, serving as APCs, supporting other mononuclear cells, and directly promoting inflammatory signal pathways.

B Cell Activation

On the one hand, B cells function as APCs to receive protein antigen activation and present protein epitopes on MHC II molecular to the helper T cells, and this mechanism is commonly referred to as T cell-dependent activation of B cells. On the other hand, some non-protein antigens, lipopolysaccharides, and polysaccharides serve as T-independent antigens, and these molecules activate B cells without the need for antigen processing and presentation to T cells. Hence this mechanism is called T Cell-independent activation of B cells.

B Cells Isolation

Generally, there are several approaches used for B cell isolation. Positive selection and negative selection as the two main methods. Positive selection is a method by which B cells are individually isolated and retained for downstream analysis after being targeted or labeled using a B cell isolation kit (positive selection). Negative selection is a method by which other cell types are removed first using a B cell isolation kit (negative selection), leaving B cells behind. In addition, B cell depletion is another method, where a single B cell type is depleted from a cell sample.
