Introduction of the CHO Stable Cell Lines

With the rapid development of protein biopharmaceuticals, especially these therapeutic recombinant monoclonal antibodies, a stable protein expression system is of great significance for biotechnological manufacture. Different from the transient gene expression system, target DNAs of stable cell lines expression system are usually integrated into the chromosomes of host cells, which can be expressed and passed on with cell proliferation. Stable mammalian cell lines are preferred for protein production with unique advantages. Currently, the most extensively and frequently used mammalian cells in research or industrial are human embryonic kidney 293 cells (HEK-293) and Chinese Hamster Ovary (CHO) cell lines. Biologics expressed in CHO cells approved in the United States and Europe accounted for about 80% of currently approved therapeutic proteins.

One of the remarkable advantages of utilizing CHO cell lines for therapeutic protein production is the similarity to the human cell system, which means lower immunogenicity and better compatibility for human disease therapy. Additionally, CHO stable cell lines offer high-yield and high-quality protein production in persistent and stable manners. Deficiently, protein expression in CHO stable cell lines is relatively time-/cost-consuming due to resistance selection and amplification, but these limitations have been improved by novel biotechnologies recently.

CHO Stable Cell Lines for Manufacturability Assessment

To develop a commercial biotherapeutic protein, two critical aspects of the candidates should be firstly considered that are expression titer and purification yield. CHO stable cell lines are preferred for the productivity assessment since they provide gram quantities of protein production with consistent quality.

Furthermore, besides CHO productivity assessment, manufacturability assessment also involves many other experimental evaluations, including physical stability assessment, chemical stability tests and post-translational modification assessment, and pre-formulation research, all of which require a large number of high-purity proteins. CHO stable cell line is the preferred expression system which can provide a large number of stable protein candidates source for various detailed characterizations and manufacturability assessments.
