Genetic-based Immunization

DNA immunization, also known as genetic immunization, is a technique for presenting the interested antigen by injecting genetically engineered DNA. Through appropriate approach, the antigen-encoding gene can be transfected into the host cells resulting in direct in vivo expression of the interested protein or peptide. A typical DNA immunization strategy will only request the information of amino acid or nucleotide sequence of the interested antigen, thereby avoiding the production and purification processes and other potential risks of contamination that could be a real challenge for certain antibody development project. In addition, the in vivo expressed antigen also takes advantages of its naïve conformation and appropriate post-transcriptional modifications (due to the host species). In general, DNA immunization is now considered as a qualified substitute for direct protein/peptide injection method, especially for developing novel antibodies against the naïve protein or for in vivo diagnostic purposes.

Polyclonal antibodies (pAbs) represent a population of antibodies collected from multiple B cell clones. pAbs recognize multiple epitopes on any one antigen, and the serum obtained contains a number of different antibodies of different affinity.

As a series of antibodies that recognize various epitopes of one specific antigen with different specificities and affinities, polyclonal antibodies are broadly used in a variety of research purposes, such as ELISA, disease diagnosis, and western blot. A large quantity of polyclonal antibodies can be generated within a shorter period and relatively lower cost than monoclonal antibodies. Compared with monoclonal antibody, polyclonal antibody can be a better choice in some specific applications (e.g. IP, WB, and ELISA). Due to the higher overall affinity and the multiple epitopes recognition, polyclonal antibodies are ideal tools that can efficiently detect or capture the target antigen, especially for samples that contain low amounts of antigen. Furthermore, polyclonal antibody production is a simpler and universal method which can be used to almost of species that can produce an immune response.
