Introduction of CHI3L1

Chitinase-3-like protein 1 (CHI3L1), also known as YKL-40, is a 39 kDa secreted glycoprotein that plays a pivotal role in the body's inflammatory and tissue remodeling responses. Produced by cells such as activated macrophages and neutrophils, its presence is a significant indicator of an underlying biological process. Unlike true chitinases, CHI3L1 lacks enzymatic activity, functioning instead as a non-catalytic lectin that regulates cell proliferation, adhesion, and migration, making it a powerful target for analysis.

Studies have extensively demonstrated the clinical relevance of CHI3L1, revealing that its circulating levels are frequently elevated in a wide range of diseases. This includes many malignancies, such as glioblastoma, as well as cancers of the colon, breast, prostate, and kidney. Furthermore, its expression is a reliable marker for pathological conditions like fibrosis, with studies showing its ability to differentiate between early and late stages of liver fibrosis. CHI3L1 is also considered a potential biomarker for cystic fibrosis disease severity, highlighting its broad utility.

Applications

Oncology

CHI3L1 serves as a crucial biomarker for tumor progression and metastasis, offering critical insights into disease aggressiveness. Its analysis can help predict patient prognosis in various cancers and, notably, assess the potential response to targeted therapies and emerging immunotherapies. This provides a valuable tool for patient stratification and therapeutic guidance.

Inflammatory Diseases

In chronic inflammatory conditions, such as liver fibrosis, rheumatoid arthritis, and cystic fibrosis, CHI3L1 analysis provides a non-invasive and precise method for monitoring disease progression, evaluating the effectiveness of treatments, and guiding personalized therapeutic strategies. This enables clinicians to better track disease activity and optimize patient care.

Neurodegenerative Disorders

The protein's role in neuroinflammation makes it a valuable biomarker for diseases like Multiple Sclerosis (MS) and Alzheimer's disease (AD). It allows for the non-invasive and real-time monitoring of ongoing glial activation in the central nervous system, offering a crucial window into disease activity.

Service Highlights

FAQs

  1. Can you analyze CHI3L1 expression in specific cell types within a tissue sample?

Yes, we are able to analyze CHI3L1 expression in specific cell types. We utilize advanced IHC techniques combined with multiplexed staining to visualize and quantify CHI3L1 protein localization within the context of a tissue sample, allowing for precise cellular-level analysis.

  2. Do you offer services to study the functional effects of CHI3L1?

We do offer services to study the functional effects of CHI3L1. These services include our specialized CHI3L1-knockdown cell line generation service for in vitro assays and the evaluation of its biological effects on cancer cell proliferation and apoptosis in a controlled laboratory setting.

  3. What are the starting material requirements for tissue analysis?

The starting material requirements for tissue analysis include properly fixed and embedded tissue blocks. We accept formalin-fixed paraffin-embedded (FFPE) tissue blocks, as these are the preferred format for our IHC and other tissue-based CHI3L1 expression studies.

  4. Can I combine CHI3L1 analysis with other biomarker services?

You can certainly combine CHI3L1 analysis with other biomarker services. We can perform multiplexed analysis that allows for the simultaneous detection and quantification of multiple biomarkers within a single sample, providing you with a more comprehensive overview of your research subject.

  5. What kind of data will I receive in the final report?

The final report you will receive will contain a comprehensive set of data. It will include all raw data from the analysis, detailed protocols and methodologies used during the assays, and a professional summary of the results with a clear interpretation of the findings for your project.
