γ-aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the mammalian brain where it acts at GABAA receptors, which are ligand-gated chloride channels. Chloride conductance of these channels can be modulated by agents such as benzodiazepines that bind to the GABAA receptor. At least 16 distinct subunits of GABAA receptors have been identified. The GABAA receptor is the site of action for many important drugs such as barbiturates, benzodiazepines, and ethanol. Among them, the GABRA6 receptor is a potential biomarker for neuronal toxicity.

A previous study found that the T1521C variant in the GABRA6 gene is related to specific personality characteristics and a significant reduction in the response of hormones and blood pressure to psychological stress. Another study indicated that a GABRA6 mutation, R46W, was identified as a susceptibility gene that may contribute to the pathogenesis of childhood absence epilepsy (CAE). CAE-associated GABRA6 R46W mutation could cause neuronal disinhibition and thus increase susceptibility to generalized seizures through a reduction of αβγ and αβδ receptor function and expression.
