Vitamin D-binding protein (VDBP), also known as gc-globulin (group-specific component), is a protein in humans encoded by the GC gene. VDBP belongs to the albumin gene family, together with human serum albumin and alpha-fetoprotein. It is a multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid and on the surface of many cell types. It is able to bind the various forms of vitamin D including ergocalciferol (vitamin D2) and cholecalciferol (vitamin D3), the 25-hydroxylated forms (calcifediol), and the active hormonal product, 1, 25-dihydroxyvitamin D (calcitriol).

VDBP Marker of Chronic Obstructive Pulmonary Disease

VDBP presents in bronchoalveolar lavage (BAL) fluid from subjects with chronic obstructive pulmonary disease (COPD) and asymptomatic smokers. In general, VDBP levels were higher in COPD. It should be noted that since transcription of GC is affected by proinflammatory cytokines, VDBP is considered an acute phase reactant. VDBP also interacts with a key mediator of lung damage in COPD-neutrophil elastase (NE) cleaving the VDBP-binding site on neutrophils. Furthermore, the ability of VDBP complexes to mediate neutrophil chemotaxis is prevented by NE inhibitors. VDBP is of great potential for diagnosis of chronic obstructive pulmonary disease.

Tuberculosis (TB) is an infectious disease usually caused by the bacterium mycobacterium tuberculosis (MTB). The disease generally affects the lungs, but can also affect other parts of the body. Given that VDBP is involved in both neutrophil chemotaxis and macrophage activation, it is recognized that airway defense against infection could be altered by GC genotype. VDBP can be a marker to elevate the mycobacterial infections, which can be involved in the acute inflammatory response and play a role in pathogenesis.
