Death-associated Protein Kinase (DAPKinase)

DAPKinase, also known as DAP-2, is a novel calcium/calmodulin-dependent serine/threonine kinase that plays an integral role in metastasis and apoptosis. DAPKinase is a 160kDa protein with an interesting multi-domain structure, including ankyrin repeats and the death domain. Located at N-terminus of DAPKinase, the catalytic domain consists of 11 typical subdomains of Ser/Thr kinases. Except for a small portion of the catalytic cassette, the remaining 80% of the DAPKinase is involved in localization and regulation. The cytoskeletal interacting domain contains a series of eight ankyrin repeats which plays an important role in the proper localization of the kinase to the actin stress fibers and for its cellular effects. As a novel protein, its expression is essential in gamma interferon-induced apoptosis. Moreover, DAPKinase activity is required for the induction of cell death according to multiple death signals which includes those generated by death receptors, matrix detachment, cytokines, and hyperproliferation. Overexpression of DAPKinase has been demonstrated to induce apoptosis in various cell lines. By contrast, the expression of DAPKinase is decreased or absent in various cancer cell lines.

DAPKinase Marker of Ovarian Cancer

Ovarian cancer is the leading cause of gynecological cancer death in the world. However, there are no effective diagnosis strategies for ovarian cancer. It has been shown that DAPKinase might be a potential biomarker for the diagnosis of ovarian cancer. It has been reported that the expression of DAPKinase is decreased or absent in various cancer cell lines. Some studies also suggest that the methylation of DAPKinase is a frequent event in ovarian cancer. Furthermore, DAPKinase inhibition limited growth of p53-mutant ovarian cancer. In summary, the lack of expression of DAPKinase along with hypermethylation relates to the ovarian carcinogenesis.
