Tau Oligomers

Tau protein is a member of the microtubule-associated protein (MAPs) family, which plays a fundamental role in the assembly and stabilization of microtubules, as well as on axonal transport and neurite outgrowth. Neurodegenerative diseases are a leading cause of death and disability affecting millions of old people. Tau protein plays an important role in the maintenance of neuronal polarity and in the stabilization of the morphology of differentiated neurons. Neurofibrillary tangles are composed of intracellular aggregates of tau protein and are key neuropathological features of Alzheimer’s disease and other neurodegenerative diseases, collectively referred to as tauopathies. Tauopathies include Alzheimer’s disease (AD), frontotemporal dementia (FTD), Pick’s disease, progressive supranuclear palsy (PSP), variants of Parkinson’s disease (PD) corticobasal degeneration, and Lewy body dementia. Tau research has become one of the central players in the investigation of neurodegenerative diseases.

Tau Oligomers Marker of Neurological Disease

Tau oligomers are one of the neuropathological hallmarks of AD and other tauopathies. Regarding NFT accumulation in AD, there is evidence showing that progressive neuronal loss and cognitive impairment correlates with the accumulation of soluble species of tau in NFTs in AD mouse models. This pathological feature has become a potential therapeutic target for the AD, as well as a diagnostic and monitoring marker of disease progression.

Tau is an intracellular microtubule-associated protein. The mechanism of tau transmission from cell to cell is still unknown. Although we do not know the exact form of toxic tau oligomers, there is increasing evidence of the possibility of tau oligomer propagation. Research focusing on extracellular tau will open potential new avenues for discovering the mechanism of tau propagation.
