miR-200b

MiR-200b belongs to the miR-200 family, which is composed of miR-200a, miR-200b, miR-200c, miR-141 and miR-429. These five members are located in two clusters, of which miR-200a/b/429 cluster is on chromosome 1p36 and the miR-200c/141 cluster is on chromosomal 12p13. miR-200 family plays a pivotal role in tumorigenesis. It has been observed that miR-200 family is downregulated in many carcinomas, such as lung, ovarian, breast cancer. Emerging studies have shown that the miR-200 family functions as tumor suppressors by regulating EMT (epithelial-mesenchymal transition), which is essential for tumor invasion and metastasis. Notably, miR-200b is the fundamental regulator in EMT beyond the other members of miR-200 family.

Regulation of miR-200 Family

The role of EMT in the progression of cancer is associated with miR-200 family by targeting ZEB1/2, which represses E-cadherin expression. It was found that overexpression of miR-200 prevents TGF-β induced ECM, in contrast, inhibition of miR-200 is sufficient to induce EMT. There has been a strong relation between ZEB1/2 and TGF-β and it was suggested that ZEB1/2 is crucial for TGF-β-induced EMT. The miR-200 family and ZEB1/2 are associated in a double-negative feedback loop, which can regulate EMT in tumorigenesis. In one side, miR-200 family target ZEB1/2 and suppress it to promote epithelial differentiation. On the other side, downregulation of ZEB1 can enhance miR-200. And this loop has a great effect on the stabilization of cellular differentiation.

miR-200b as a Diagnostic Marker for Cholangiocarcinoma

Several studies have shown that miR-200b is downregulated in some types of cancers including breast cancer, and it was suggested as a prognostic factor in breast cancer. Similarly, in Cholangiocarcinoma (CCA), it was found that four miR-200 members, including miR-200a/b/c429 are decreased. Furthermore, upregulated miR-200b/c prevents the migration and invasion of CCA cells. The mechanism that miR-200b/c influences the CCA tumorigenesis is involved the tumor-initiating capacity, sphere formation and drug resistance by directly targeting rho-kinase 2 and SUZ12. Hence, miR-200b also is an important prognostic biomarker for CCA.
