Background

L1CAM, a transmembrane glycoprotein of 200-220 KDa, is a neuronal cell adhesion molecule belonging to the immunoglobulin superfamily. L1CAM plays key functions in the development of the nervous system including neuronal migration, differentiation and is also involved in the malignant progression of neoplasia and the metastatic cascade.

In cancer cells, L1CAM promotes cell proliferation, migration, invasion, and metastasis. Overexpression of L1CAM has been reported to be strongly associated with numerous human cancers such as colorectal, gastric, renal, breast cancer, melanoma and endometrial cancer. These indicate that L1CAM is a candidate serum biomarker for some tumors. Variants in the L1CAM gene result in a spectrum of allelic disorders with X-linked inheritance.

Scientific Validity

The extracellular domain of L1CAM can be cleaved from the tumor cell surface by shedding. The process of L1CAM ectodomain shedding enhances cancer cell migration in various extracellular matrix components through autocrine or paracrine binding of integrins. Thus, L1CAM can also be detected as soluble form of L1CAM in the serum and ascites. Besides, studies revealed that circulating autoantibodies against L1CAM could be also used as potential biomarkers for early detection of ESCC.
