Background

The gene encoding alpha-1 antitrypsin (AAT) is SERPINA1 which consists of seven exons and six introns. After translation, transcription, and post-translational modifications (PTMs), SERPINA1 is released into the circulation. As a known genetic cause of chronic obstructive pulmonary disease (COPD), alpha1-antitrypsin deficiency (AATD), SERPINA1 had the possibility to be used as a biomarker to provide diagnostic and prognostic information.

Diagnostic Power

As an endogenous anti-inflammatory protein, SERPINA1 has its own advantages over other immunotherapy candidates. SERPINA1 is an acute phase reactant secreted mainly by hepatocytes during inflammation. Its dominant function is to inhibit elastase released by neutrophils. Besides, SERPINA1 counteracts the effects of inflammatory mediators, exhibits anti-inflammatory as well as immunomodulatory effects that can be independent of protease inhibition. Many anti-inflammatory agents would inhibit appropriate immune response, while SERPINA1 does not.
