Background

VCAM1, also known as CD106, is an integrin and Ig superfamily group member that is induced on endothelial cells in response to numerous inflammatory cytokines such as TNF, IL1, and bind integrin partners on leukocytes. VCAM1 is a transmembrane type 1 protein expressed in several species and has different conformations. As Fig.1 shown, VCAM1 contains an extracellular N-terminal portion, with a variable Ig-like domain produced by splice variants of the VCAM1 gene. There are two isoforms of human VCAM1: one with 7 Ig-like domains (full length) and one with 6 Ig-like domains (with excision of domain 4).

Scientific Validity

VCAM1 expression, together with soluble VCAM1 (sVCAM1) induced by the shedding of VCAM1 by ADAM metallopeptidase domain 17 (ADAM17) have been proposed as biomarkers in various disease such as cardiovascular disease, immunological diseases, cancer and autoimmune myocarditis. The protein is cleaved near the transmembrane site. Several studies have shown elevated and strongly correlated serum and urinary VCAM1 levels with lupus nephritis activity and severity.
