Introduction to Antibody Fc Region

Fc region is a crystallizable fragment of an antibody that located at the base of the "Y", which is responsible for binding to Fc receptors. The Fc region consists of two heavy chains that contribute two or three constant domains depending on the isotype of the antibody. There are five different types of Fc regions in general, which allow antibodies to be grouped into five isotypes that are IgG, IgM, IgA, IgD, and IgE. Exception for IgD, each Fc region of a certain antibody isotype binds to corresponding Fc receptor and enables the antigen-antibody complex to exert different roles depending on which Fc receptor it binds.

By binding to specific Fc receptors and other immune molecules, the Fc region ensures that a specific antibody generates an appropriate immune response for a different antigen, and thus mediating different physiological effects and effector functions, including but not limited to:

  • Recognition of opsonized particles when binding to Fcγ receptors
  • Degranulation of mast cells, basophils, and eosinophils when binding to Fcε receptors
  • Lysis of cells or complement-dependent cytotoxicity when binding to complement proteins
  • Antibody-dependent cellular cytotoxicity (ADCC)
  • Antibody-dependent cell phagocytosis (ADCP)
  • Interaction with the neonatal Fc receptor (FcRn) to slow down antibody degradation and extend its serum half-life
