Importance of Glycosylation

Glycosylation refers to the enzymatic process that attaches glycans to proteins, or other organic molecules. As a form of co-translational and post-translational modification, glycosylation serves a variety of structural and functional roles in both membrane-associated and secreted proteins. There are five classes of glycans which are N-linked glycans, O-linked glycans, phosphoglycans, C-linked glycans, and glypiation. Glycosylation is important in biology because it affects the way that proteins or lipids behave in the body and how they interact with other molecules. What’s more, it is also important in the development of anti-cancer medicines.

Fully Human Glycosylation Services

Glycosylation is important for the efficacy, pharmacokinetics, and immunogenicity of therapeutic antibodies. The glycosylation patterns vary in terms of monosaccharide composition, glycosidic linkages, and glycan branching. The host cell line used to produce therapeutic antibodies has a strong influence on the glycosylation because different host systems may express a varying repertoire of glycosylation enzymes and transporters that contribute to specificity and heterogeneity in glycosylation profiles. Most of the marketed biologics are derived from non-human cells that are genetically engineered to express human-like glycosylation patterns.

Human cell lines are a promising and emerging alternative to non-human host cells as the resultant recombinant antibodies would possess fully human glycosylation patterns, reducing the cost for downstream processing and, more importantly, circumventing the risk of immunogenic reactions from non-human glycans.
