The identification and validation of disease-causing target genes is an essential first step in drug discovery and development. In contrast to gene-profiling approaches, the loss-of-function approach is playing a big role in drug discovery and development, especially when in term of target identification. It provides the information for the direct link between gene alteration and the disease phenotype and offers strong proof or disproof for a certain mechanism of action. Homologous recombination is a reciprocal exchange of DNA sequences, as in between two chromosomes that carry the same genetic loci. It is conserved and occurs in diversified organisms, but all forms basically employ the same steps. More importantly, homologous recombination is valuable for generating mutation. When a double-strand break occurs, sections of DNA around the 5' ends of the breaks are cut away in a process called resection. In the ensuing step, an overhanging 3' end of the broken DNA molecule “attacks” a similar or even identical DNA strand that is not broken. Then, the repair machinery in the cell would be induced. Homologous recombination is widely utilized to accurately repair breaks that occur on both DNA strands. By taking advantage of the mechanism, the repair machinery of double-strand break is employed in our platform to generate mutant cells. After that, a series of assays would be carried out. Homologous recombination is an alternative gene knockout platform of Agrobacterium tumefaciens-mediated transformation which inserts mutagenesis or transposon to create mutagenesis. Our gene targeting platform has been successfully used in traditional and conditional genetic modification research.
