The Oris cell migration assay is based on a multi-step process, in which a mask is applied to the bottom of a 96-well microplate well first. Meanwhile, a polymeric insert (silicon stopper) is installed in the wells of the microplate. When cells are added, the insert prevents cells from seeding the internal analysis area. After cell attachment, the insert is removed, allowing cells to migrate freely to the central analysis area of the wells. Cells that do not migrate to the analysis area will not be observed due to the placement of the mask originally at the bottom of the plate. Migrating cells were analyzed by a microscopy or a synergy HT multimode reader with fluorescence.
