Plaque reduction neutralization assay (PRNT) is a vital method used to measure the ability of antibodies to neutralize cells infected with virus particles. It is considered to be the most virus-specific serological test currently and is widely used in research.

Commonly, infectious virus particles multiply in cells, forming circular areas of the infected regions, known as plaques. In vitro plaque reduction assay evaluates the neutralization ability of antibody samples by measuring the plaque forming efficiency of a virus in the presence of antibody samples at different concentrations.
