What Is A Knock-out Mouse Model?

Knock-out mice models are defined as models that have been genetically modified to disrupt or inactivate one or more specific genes. These alterations in the genome will cause a change in the animal model phenotype. Mouse models are extensively used in the in vivo studies of gene function, human disease research and the development of novel drugs, due to a high similarity they share with humans in the genome. As a result, by observing the characteristics of knock-out phenotype, investigators are able to get a better understanding of human gene function and genetic diseases.

Conventional (constitutive) Knock-out Mice

This model is engineered to carry a permanently inactivated target gene in every cell of the organism at all times. Generally, the inactivation can be realized by partial or total deletion of the sequence of the gene of interest.

Conditional Knock-out Mice

This model is characterized by inactivating a specific gene at specific times during development or in specific tissues rather than being deleted from the beginning of life. It is a better choice when the conventional knock-out is predicted to cause lethal effects to the mouse model.

Tissue-Specific Knock-out Mice (conditional)

This model is also a conditional type which inactivates gene expression in a specific target tissue, thus circumventing the lethal phenotype caused by conventional mutants. This type of model is often used to study the function of lethal genes, identify target gene of drugs, and evaluate drugs in preclinical studies.

Inducible Knock-out Mice

This is a conditional biological model in which the expression of the target gene can be specifically inactivated in a time-controlled manner. Thus, the model expresses wild-type proteins and mutant proteins before and after induction, respectively. Especially, this model can be used in studies of diseases that are related to development or age.

KO First (Conditional Ready) Models

KO First, is a multipurpose mouse model which use a modified conditional knock-out strategy to first obtain the whole-gene knock-out mouse model, and then obtain the conditional knock-out mouse model after mating with a proper mouse model.
