Introduction of Luminescent Cell Viability Assay

The luminescent cell viability assay is a highly sensitive homogenous method to quantify the ATP present. in cell cultures. Because ATP is an indicator of metabolically active cells, the amount of ATP available can be used to assess the number of viable cells. In this assay, the luminescent signal caused by proprietary thermostable luciferase is proportional to the amount of ATP present. By correlating the amount of ATP with the number of viable cells, this assay has a wide range of applications, such as cell proliferation, cell viability, and cytotoxicity. This assay is highly sensitive to detect single cell or 0.01 picomoles of ATP, the produced signal is linear within six orders of magnitude. In past projects, our assay has been shown to detect a variety of bacteria and fungi.

Luminescent Cell Viability Assay for MOA Study

Recent studies showed that cell membrane provides a selective barrier for the stability of bacterial cells and the conduction of energy and matter. What’s more, the cell membrane also participates in many important physiological processes. In this case, cell membranes present great potential as antimicrobial targets. The developed antimicrobial peptides (AMPs) and a series of membrane-active antibiotics have been successfully used in medicine, and their bacterial specificity also avoids damage to mammalian plasma membranes. In MOA studies of antibacterial agents, if the suspected mechanism is targeting the membrane, the luminescent cell viability assay would be used for further verification. In this case, polymyxin B would be served as the positive control.
