Introduction of Mutant Prevention Concentration (MPC)

In the history of the misuse of antibiotics, the emergence of antibiotic resistance has become an increasingly serious public health problem. Antibiotic resistance can be achieved through horizontal acquisition of resistance genes, recombination of foreign DNA into chromosomes, and mutations at different chromosomal loci. Antibiotic resistance studies can help predict the frequency of drug-resistant mutants and the types of drug-resistant strains that may appear clinically.

The mutant prevention concentration (MPC) is defined as the minimum concentration restricting the growth of the least susceptible, single-step mutant of a bacterial isolate. The drug concentration between the MIC and MPC is called mutant selection window (MSW) to avoid the bacteria growing with resistance-associated mutations and accumulating further mutations. Lower MPC/MIC ratio values indicate greater ability to prevent mutants from appearing. During the development of novel antibiotics, the MPC is a well-known concept and combining research on MPC may further limit the emergence of drug resistance and increase the efficacy of current treatments.

Procedure for MPC Determination

In this assay, MIC data were used to plan incremental concentrations used in MPC experiments and the typical concentrations can range from 1 × liquid MIC to 64 × liquid MIC. At first, starter cultures were spread on blood Trypticase soy agar plates and the plates were incubated overnight at 35 to 37°C. The cells were then recovered from the plates and inoculated on LB plates containing antibiotic dilutions. Finally, MPC can be obtained when the agar plate shows zero colonies. To ensure the accuracy and reproducibility of the assay, we will do three sets of tests.
