Vaginal mucosa, as one of the body's hosts defense and immune surveillance components, is a natural barrier that is constantly exposed to bacteria and viruses. However, the vaginal epithelium can be damaged mechanically, e.g. by the incorrect use of objects such as tampons, or by chemicals that are irritating or corrosive. Consequently, this can make the tissues more susceptible to infections. Therefore, it is essential to predict the effects of personal care, cosmetic and pharmaceutical products for vaginal use on humans before the product is marketed.

Introduction of Vaginal Mucosal Irritation Test

For a long time, the rabbit vaginal irritation test (RVI) has been the most frequently used method for assessing vaginal mucosal irritation. Although the vaginal tissues between rabbit and human are dissimilar, the rabbit method is still preferred and widely used to determine the vaginal tolerance or mildness of topical spermicidal preparations. While, in recent years, the concept of the 3Rs (refinement, replacement, and reduction) has stimulated the development of alternative in vitro methods. Various in vitro studies, such as cell lines and reconstituted tissue models, have been confirmed that they have the potential to replace the RVI model when used for cosmetic and personal care product screening.
