Cytomegalovirus retinitis, also known as CMV retinitis, is a serious viral eye infection of the retina. It is caused by human cytomegalovirus and most often found in people with weakened immune systems. Over the last years, antisense oligonucleotides (ASOs) have gained attention as a therapeutic tool for the treatment of ocular diseases such as CMV retinitis. Depending on the complementary to their target, the ASOs can result in modulation of pre-mRNA splicing, or in a (allele-specific) degradation of target transcripts. It has documented that ASO-based strategies are capable of correcting splice defects resulting from deep-intronic mutations and inducing allele-specific degradation of mutant transcripts in ocular diseases. Some antisense compounds have been designed to inhibit CMV replication, which possesses antiviral activity that favors their use as therapeutic agents in the treatment of CMV retinitis.
