It is known that retroviral vectors are retroviruses that have been genetically engineered to deliver therapeutic genes to target cells. The typical feature of retroviral vectors is that the transgenes they carry integrate into the host DNA and can potentially maintain permanent gene expression. There are many types of retroviruses that are used for the preparation of vectors, including bovine leukemia virus, Rous sarcoma virus, lentiviruses and spumavirus, etc. Among them, the most popular vectors are constructs based on Moloney murine leukemia virus (MoMLV) and HIV. Depending on the application, retroviral vectors can be constructed as replication-competent or replication-deficient. In terms of gene therapy, different retrovirus vectors have been used to carry transgenes inducing cell death and enable long-term expression of the transgene. Currently, retroviral vector-mediated gene delivery has been central to the development of gene therapy.
