Glycation Assay

Therapeutic antibody drugs have developed rapidly in recent years due to their efficacy in a wide variety of disease therapy, such as various tumors, immune system diseases, and infectious diseases. Recent studies have also revealed that protein post-translational modification (PTM) plays a critical role in regulating the structure and function of candidate drugs, especially for antibody drugs. As a consequence, Creative Biolabs provides a number of structural detection and glycosylation analysis services on drug targets, including small molecules and antibodies, to avoid the reduction of the efficacy of potential drugs.

Introduction

Glycation is a non-enzymatic chemical modification. For recombinant antibody drugs, glycation mainly occurs on residues such as lysine (Lys), and together with post-translational modifications such as glycosylation, oxidation, deamidation, and isomerization, result in high heterogeneity of recombinant antibody drugs, producing substances or impurities related to recombinant antibody drugs. Glycation may have an impact on the structure, physical and chemical properties, safety and efficacy, and stability of recombinant antibody drugs.

Our Technologies for Glycation Analysis

In the actual sample test, Creative Biolabs has developed a series of advanced technologies for testing the glycation for your targets. The Maillard reaction has been used to speculate the occurrence of protein glycation in the solution. The separation of glycated and non-glycosylated proteins can be carried out by boric acid affinity chromatography or ion exchange. Mass spectrometry can be used for the final confirmation of glycation sites. For instance, the light and heavy chains of glycated antibodies have a broader peak in capillary electrophoresis-sodium dodecyl sulfate (CE-SDS), while the light chain has a narrower peak under UV or laser induction. Therefore, a combination of CE-SDS and CE-MS techniques can be used to analyze glycation. Since glycation primarily causes acidic variants of recombinant antibodies, cation exchange chromatography techniques can be used to prepare acidic variants and further studies of antibody glycation are performed.
