This GFUS knockout (HEK293F) cell line lacks the GFUS protein, also known as Tissue Specific Transplantation Antigen P35B, a GDP-L-fucose synthase. This NADP(H)-binding protein catalyzes the two-step epimerase and reductase reactions in GDP-D-mannose metabolism, converting GDP-4-keto-6-D-deoxymannose to GDP-L-fucose. GDP-L-fucose is the substrate of several fucosyltransferases involved in the expression of many glycoconjugates, including blood group ABH antigens and developmental adhesion antigens. Loss of GFUS function may disrupt GDP-L-fucose synthesis, impacting the formation of various fucosylated glycans and potentially affecting cell signaling, adhesion, and development.
