Description GlycoFlux™ human PPP1R12A overexpression HepG2 cell line is a stable glycoengineered line that constitutively overexpresses MYPT1, the myosin phosphatase targeting subunit. By reshaping actomyosin contractility and vesicle dynamics, it provides a means to quantify how cytoskeletal tone governs Golgi stacking, cargo transit, and glycoform distribution on membrane receptors.

Summary Myosin phosphatase target subunit 1 (MBS) is a component of myosin phosphatase, regulating actin-myosin interactions downstream of the small GTPase Rho. Active RhoA binds MBS, influencing myosin light chain (MLC) phosphorylation via Rho-kinase. This phosphorylation reduces myosin phosphatase activity, promoting smooth muscle contraction and actin-myosin association in nonmuscle cells. Overexpression of RhoA or its active form increases MBS and MLC phosphorylation. Several isoforms result from alternative splicing.
