Publication

This publication addresses the challenge of generating antibodies for membrane proteins, which is often limited by the availability of purified or natively folded antigens. The study describes a genetic immunization approach that uses DNA alone to produce antibodies against a diverse set of bacterial and viral membrane proteins. They successfully produced antibodies for 71% of the 17 targets studied.

To facilitate antibody characterization, the researchers also developed a new in vitro method, called IVT-HMB, for the simultaneous expression and capture of membrane protein targets. This method simplified antigen preparation by eliminating the need to purify detergent-solubilized or urea-denatured membrane proteins. The antibodies generated from this approach were used to characterize the expression and membrane-targeting of several F. tularensis proteins, demonstrating antibody specificity. The authors conclude that these methods will accelerate future structural and functional studies of therapeutically relevant membrane proteins.
