Introduction of Cell Line Characterization

Cell line characterization is a key step in ensuring the safety of biopharmaceutical products to confirm the identity, purity, genetic stability, and tumorigenicity of cell lines. This characterization can ensure that the source of the cell line is authentic and free from contamination, undesirable cells, and foreign factors such as bacteria, fungi, mycoplasmas, mycobacteria, and viruses. International regulatory guidelines such as ICH (Q5A, Q5B, Q5D), EP, and USP for cell lines characterization mainly focus on three areas:

  1. Source history and generation of cell lines
  2. Establish characteristics and tests of cell line identity, purity, and genetic stability
  3. The banking procedures of the cell line.

How Creative Biolabs Can Help

Core steps of the cell line characterization process. (Creative Biolabs Original)

Highlights

Master Cell Integrity

We ensure the stability of your production platform by verifying the genetic consistency of the MCB and subsequent WCB. Our analysis confirms low passage numbers and maintains the single master cell line integrity.

Molecular Fingerprinting

Creative Biolabs provides definitive identity confirmation through rigorous short tandem repeat (STR) profiling and DNA fingerprinting. This legally defensible evidence guarantees the authenticity of your cell line and prevents costly cross-contamination incidents.

Service Features

Comprehensive Purity Testing

We conduct extensive purity testing, including highly sensitive screens for Mycoplasma, viruses, and fungi detection. This essential quality control step secures the long-term safety and sterility of your cell banks against critical adventitious agents.

Audit-Ready Documentation

Our documentation is built for global compliance, incorporating ICH Q2-validated assays. This guarantees your data package is scientifically robust, acceptable to regulators, and dramatically accelerates the quality review process.

FAQs

Which stages of my cell line development require full characterization, and what's the difference?

Full, exhaustive characterization is primarily required for the MCB. However, the WCB, the RCB, and cells at the limit of in vitro cell age (LIVCA) also require specific, tiered testing. LIVCA testing is crucial for demonstrating that the cell line remains genetically stable throughout the entire manufacturing lifespan.

What is the biggest precaution I should take before submitting my MCB for characterization?

The biggest precaution is ensuring your cell bank is free of mycoplasma contamination and that you have a clear, accurate record of the cell line's passage history. Mycoplasma contamination is the most common reason for project delays. We recommend pre-screening for mycoplasma using our high-sensitivity qPCR before formal CLC begins.
