Claudin 6, a 23 kDa protein, is an important component of the CLDN family and serves a key role in the functions of tight junctions. Claudin 6 shares the common structure of Claudin proteins, consisting of four transmembrane domains, two extracellular loops (ECL1, ECL2), and the intracellular NH2, COOH ends. Claudin 6 can bind with signal proteins and cytoskeletal proteins, inducing epithelial cell junction formation and polarity, regulating the cell proliferation, differentiation and apoptosis of the epithelial cell.

Claudins are involved in a variety of epithelial-derived cancers. Claudin 6 has been found to be an oncogenic gene in most malignant tumors and may be involved in tumor formation or tumor development. As Claudin 6 is widely expressed in various tumors but rarely expressed in healthy adult tissues, it is used as a promising target for a variety of therapeutic strategies, such as antibodies, ADCs, antibody-drug radionuclide-targeted drugs and CAR-T therapies.
